3-D Life is the most flexible system for biomimetic hydrogel design for 3D cell cultures. It uses two polymers that form a hydrogel by covalent crosslinking at ambient temperature. The chemical reaction is non-toxic to cells and occurs rapidly, preventing cell settlement before the gel is formed. Thiol groups on crosslinker CD-Link form stable thioether bonds with maleimide groups on Mal-Dextran, resulting in gel formation. Cell adhesion peptides (RGD Peptide, Cat. No. 09-P-001) can be attached to Mal-Dextran to provide a cell-adhesive matrix. CD-Link is designed for MMP cleavage, allowing cell spreading/migration if they express the indicated MMPs. Dextran hydrogels can be dissolved with addition of dextranase (Dextranase, Cat. No. D10-1), allowing easy cell recovery. One kit allows formation of up to 2 ml hydrogel depending on gel stiffness.
